Hamilton was born and raised in Raleigh, North Carolina, where he attended Athens Drive High School, where he excelled as an outfielder and pitcher. During his high school years, he was extremely popular, and he was selected as one of the top two players in the 1999 MBL Draft. Hamilton was signed by the Tampa Bay Devils and received a $3.96 million bonus as a result of his talent.

Later, Hamilton joined the Hudson Valley Renegades and led them to their first New York-Penn League championship. He also played for the Charleston Riverdogs in the South Atlantic League, where he was named MVP of the South Atlantic League All-Star Game. However, due to an automobile accident, injuries, and subsequent drug addiction issues, Hamilton did not play baseball again until 2007.

The Chicago Cubs picked Hamilton in the Rule 5 draft while picking for the Reds, and the Rangers purchased him for $100,000. During the 2008 season, he was named to the All-Star team, and he hit 28 home runs while competing in the Home Run Derby, finishing with 35 runs, making him only the second player in history to do it.

He also won the AL batting title in 2010 and was selected as the MVP of the 2010 ALCS later that year. In November 2010, he was named the 2010 American League MVP. He also moved up to 16th place on the list of only players in MLB history to blast four home runs in a single game.

Hamilton was selected player of the month shortly after being dealt to the Texas Rangers. He also won Player of the Month for the second month in a row, making him the first player in American League history to win Player of the Month twice in a row within the first two months of the season. He was then featured on the cover of Sports Illustrated in 2008. On July 9, 2008, Hamilton hit his first walk–off home run.
